C1 PURIFIED HUMAN COMPLEMENT PROTEIN

SOURCE MATERIAL: Human Serum/Plasma.

BUFFER: 5 mM Barbital, 143 mM NaCl, 0.5 mM MgCl2, 0.15 mM CaCl2, pH 7.2

PROTEIN CONCENTRATION: Total Protein: 0.50 – 5.0 mg/mL using an extinction coefficient of E1%/280 nm = 8.9 C1 protein: >0.19 mg/mL based on functional activity measurements.

FUNCTIONAL ACTIVITY*: >2.5 X 105 C1H50 U/mg >70% of C1 activity in NHS on a mg/mg basis > 5 x 104C1H50 U/vial

IMMUNOCHEMISTRY: < trace amounts of C3, C4, or C5

PRESERVATIVE: None, filtered through a 0.22 m pore size filter

DISEASE STATE TESTING: Starting material is tested and found negative for HIV 1 and 2 antibodies, Hepatitis B surface antigen, HCV antibodies and HIV-1 antigen.

STORAGE: -70°C or Below. Avoid repeat Freeze/Thaw

* One C1H50 unit is defined as the amount of C1 required to yield 50% lysis of 3 x 107 EA when incubated in the presence of 1 m g C4 and 0.2 m g C2 at 30° C for 12 minutes in a total reaction volume of 500 m l SGVB++. The reaction is developed by addition of 1 ml of guinea pig serum diluted 1:50 in GVB0, 40 mM EDTA, pH 7.21, for 30 minutes at 37° C.
